苜蓿抗盐愈伤组织的诱导及其生理生化指标的测定 被引量:15

摘要 对苜蓿愈伤组织用较高浓度的盐(0.8%NaCl)进行胁迫,并用NaN3进行诱变处理,再用高盐浓度的培养基(含 NaCl1.2%以上)进行筛选,筛选后对其抗盐性进行测定。经过脯氨酸含量、过氧化物酶(POD)活性、细胞膜相对透性的 测定后,结果表明:此愈伤组织在含1.2%的NaCl培养基上长势很好,各项生理指标明显高于未处理的,具有很强的耐 盐性。 Alfalfa calluses was stressed with 0.8% NaCl, and mutagenized with NaN 3, and then mutants were screened from the medium with high concentration salt NaCl(1.2%). Finally, proline content, peroxidase activity (POD)and relative cell memberane peameability, related with salt-resistant characteristics of mutants were measured. The result showed that acquired mutants of callus grew well in the medium including 1.2% NaCl concentration. Proline content, POD and relative cell memberane peameability of the mutants were better than those of control. The mutants had a better salt-resistant characteristics.
