鼠脑缺血后再灌注c-fos基因表达及银杏黄酮的影响 被引量:7

摘要 目的 观察大鼠脑缺血再灌注后c fos基因表达、细胞凋亡及银杏黄酮对它们的影响。方法 选择健康Wis tar大鼠 5 6只 ,雌雄各半 ,鼠龄 80~ 12 0d。用大脑中动脉栓塞 (MCAO)再灌注模型 ,用原位杂交与免疫组化方法检测c fosmRNA及蛋白表达 ,应用TUNEL染色方法观察细胞凋亡。结果 银杏黄酮可明显减轻缺血再灌注后脑损伤及细胞凋亡的发生 ,可明显增强c fosmRNA及c fos蛋白表达。结论 银杏黄酮可增强并延长c fos蛋白表达 ,这可能是其脑保护作用机制之一。 Objective To study the effects of extract of Ginkgo biloba(EGB) on the expression of c-fos gene and apoptosis in rat brain after ischemia and reperfusion.Methods The animal model of focal cerebral ischmia and reperfusion was adopted,and treated with EGB.The expression of c-fos protein was assessed using immunohistochemical method.In situ hybridization and in situ end labeling methods were used to detect the c-fos mRNA and neuron cell cell apoptosis respectively.Results EGB can reduce reperfusion injury and cell apoptosis significantly and increase the expression of c-fos protein and mRNA.Conclusion EGB may play a role in protection of ischemic nenrons by increasing expression of immediate early gene c-fos.
