哮喘大鼠T辅助细胞功能异常与嗜酸细胞凋亡 被引量:1

摘要 为观察哮喘大鼠T辅助细胞 (helperTlymphocyte ,TH) TH1/TH2平衡及嗜酸细胞 (eosinophil,EOS)凋亡率的变化并探讨两者之间的关系。 2 0只清洁级雄性SD大鼠随机分为正常对照组 (C) ,哮喘组 (A) ,每组 10只。卵清白蛋白复制大鼠哮喘模型 ,ELISA法检测血及支气管肺泡灌洗液 (BALF)中IL 4、IFN γ水平 ;TUNEL法检测大鼠气道壁EOS凋亡率。结果显示A组大鼠BALF中IFN γ水平较C组明显降低 (P <0 0 0 1) ;而A组血及BALF中的IL 4水平则均高于C组 (均P <0 0 0 1) ,表现出明显的Th1/Th2失衡。A组大鼠气道壁的EOS浸润明显增多 ,但凋亡细胞却很少看到 ,EOS凋亡率为 (5 2 8± 2 2 1) %明显低于C组 (15 88± 2 39) % (P <0 0 0 1)。以上研究表明哮喘大鼠在气道壁EOS凋亡减少的同时表现为明显的Th1/Th2失衡。 To investigate the change of helper T lymphocyte (TH)- TH1/TH2 balance and eosinophil apoptosis in asthmatic rat and to identify the relationship between them. Twenty rague-Dawley(SD) rats were randomly divided into 2 groups: control (C), asthma (A). Rats in group A were intraperitioneally sensitized and nebulizedly challenged with ovalbumin(OVA) and those in group C were treated with saline instead of OVA. The assessment of IL-4, IFN-γ in both serum and bronchoalveolar lavage fluid(BALF) was performed. The Eosinophil apoptosis in airway of rats was also measured. There was a distinct imbalance of Th1/Th2 in group A. The production of IL-4 both in serum or in BALF in groups A were significantly increased from the baseline ( P<0.01) and IFN-γ production in BALF was greatly lowered(P<0.01), compared with that in group C. In group A, the number of EOSs presenting in airway of rats increased greatly. But, there were few apoptotic EOSs; the EOS apoptotic rate was (5.38+2.21)% in group A, while that in group C was (15.88+2.39)%(P<0.01). This study indicates that the decreased apoptosis rate of EOSs in airway in asthmatic rat is combined with a distinct imbalance of Th1/Th2.
