摘要
本实验采用cDNA芯片研究D-半乳糖皮下注射小鼠海马的基因表达变化。小鼠皮下注射D-半乳糖[5 0mg/ (kg.d) ]6 0d造模 ,Morris水迷宫测试小鼠行为 ,分别提取模型组和对照组海马组织总RNA ,掺入荧光分子Cy3和Cy5 ,经逆转录合成cDNA荧光探针与 4 0 0 0点小鼠cDNA基因芯片杂交。结果显示 ,在 4 0 0 0条待研究的基因中 ,两组小鼠海马间存在 2倍表达差异的有 76条 ,其中上调 2 6条 ,下调 5 0条。经分析 ,模型组基因表达谱与老年性痴呆 (AD)相关基因表达谱在氧应激、能量代谢相关基因等方面有相似之处。提示D
To investigate the differentially expressed genes of D-galactose Induced dementia model mice, D-galactose( 50 mgkg -1) was subcuticularly injected for 60 days. The learning-memory ability of the model group and the control group was tested by Morris water maze experiment. Both groups′ mRNAs were reversely transcribed to cDNA by preparation of the hybridization probes using Cy3-dUTP and Cy5-dUTP. The mixed probes were hybridized to the cDNA microarray. The cDNA microarray was scanned for fluorescent signals and showed differences between the model group and the control group. Among the 4 000 genes, expression level of 76 genes differing in D-galactose induced dementia mice among which the 26 target genes up-regulated and 50 target genes down-regulated. The gene expression in hippocampal tissue of D-galactose induced mice was related to oxygen stress, energy-metabolizing, cell apoptotosis and microtubule construction, and the regulation of gene expression was similar to that of Alzheimer disease. D-galactose mouse model may be taken as a kind of Alzheimer disease-simulating model.
