摘要
对假丝酵母 99 12 5脂肪酶的发酵工艺条件进行了一系列研究。选择了合适的培养基成分并进行优化 ,获得了最优的摇瓶培养基配方 (% ,W V) :豆油 4 0 ,全脂豆粉 4 0 ,K2 HPO4 0 1,KH2 PO4 0 1。产酶水平能达到 5 0 0 0IU/mL。在 30L发酵罐上进行初步放大实验 ,其产酶水平能达到 810 0IU/mL。在 1m3发酵罐上进行中试放大 ,产酶水平可达到 80 0 0IU mL。
Lipase catalyses many ester synthesis and transesterification reaction. The lipase from Candida sp. has good esterification ability without positional specificity for triglycerides and has potential application in the synthesis of short chain esters as flavor compounds or biofuels such as biodiesel. The fermentation of lipase with Candida sp.99-125 has been studied. The effects of different carbon resources and nitrogen resources on the lipase production were investigated. It was found that soybean oil and soy powder can enhance the lipase production. The optimal medium composition is 4% soybean oil, 4% soy powder, 0.1% K 2HPO 4 and 0.1% KH 2PO 4. The suitable shake flask fermentation conditions was identified and the lipase activity in shake flask reached 5000 IU/mL. The process was scaled up from a laboratory bioreactor (30L) to a pilot fermentor (1m 3). The amount of lipase reached 8100 IU/mL in 30L fermentor and 8000 IU/mL in 1m 3 fermentor.
