摘要
对带有烟草环斑病毒(TRSV)的淡黄花白合(Lilium sulphureum Baker)珠芽生长点进行组培脱毒。经试验得出,珠芽生长点诱导成愈伤组织的最适培养基为MS+BA0.5+2,4-D0.25mg/l,诱导率为92.8%。适合幼芽分化的培养基为MS+BA1.5+NAA0.1+KT0.1mg/l,分化率为71.4%。适合生根的培养基为1/2MS+IBA0.25mg/l,生根率为100%。从外植体到发育成幼苗约需120~150天。将培养出的幼苗在6种指示植物上进行脱毒鉴定,结果表明,0.3~0.8mm长的珠芽生长点其诱导的幼苗成功地脱去了百合的烟草环斑病毒。
The growing points in bulblets of Lilium sulphureum Baker with tobacco ringspot virus (TRSV) were used for devirus by means of tissue cultue.This experiment showed that the most suitable medium on which callus was induced from the growing points of the bulblets was MS+BA0.5+2,4-D0.25mg/ 1 and the inductivity was 92.8%.The medium suitable for bud differentiation was MS+BA1.5+NAA 0.1+KT0.lmg/ 1 and the rate of differentiation was 71.4%.l / 2MS+IBA0.25mg/ 1 was fit for root growing and the multiplication rate was 100% . It took 120~ 150 days to develop plantlets from the explants of Lilium sulphureum Baker. These plantlets were used for devirus identification by six indicator plants. The results showed that the TRSV was successfully eliminated in the plantlets induced from the growing points of the bulblets 0.3 ~ 0.8mm long.
