摘要
对饥饿及饱血银盾革蜱雌虫脂肪体的扫描电镜观察表明:饱血时,其脂肪体体积显著增大,约为饥饿时的8-10倍。随着交配及饱血,脂肪体营养细胞超微结构发生了一系列变化。交配前细胞内仅有少量分散的粗面内质网,高尔基复合体及糖原颗粒很少。交配、饱血后,细胞内含有大量平行的粗面内质网,且其小池逐渐膨大。脱落至产卵前,细胞呈现出蛋白质合成活跃状态,产卵末期,细胞逐渐趋于衰老死亡。SDS-PAGE结果表明:饱血并未使雄虫可溶性蛋白发生明显变化。雌虫在饥饿及交配前,也未出现明显的蛋白质合成增加。交配后,蛋白带大大增加,其中分子量为67kD,55kD,50kD的三种蛋白在各个时期变化显著。
The fine structure of the fat body of Dermacentor niveus Neumann in various physiological phases was studied by scanning and transmission electron microscope. The subunits of the soluble proteins of the fat body were analysed by SDS-Polyacrylamide gel electrophoresis. It indicated that the volume of fat body of the fed adult females was found to be 8-10 times larger than that of the unfed adult female ticks. With feeding and mating,the ultrastructure of fat body in adult females underwent a dramatical change. In unfed ticks, the cells of fat body were only filled with a few scattered RER, Golgi bodies and glycogen particles. After mating and feeding,the amount of RER increased distinctly.The cisternae of RER appeared gradually distended.The glycogen particles and mitochondria were often seen.After detachment from the host,the cells of fat body appeared as a kind of cells having some characteristics that were connected with active synthesis of proteins.After oviposition,the cells turned to be exhausted cells that were near death.In SDS-PAGE,here were no obvious differences between unfed and fed adult males. In female, unfed and unmated adults showed inactive protein synthesis.After mating,the bands of proteins increased greatly in adult females. From mating to oviposition, the content of three kinds of protein with molecular weight of 67kD, 55kD, 50kD was different. These proteins may have something to do with the development of ovary and eggs.
