内蒙古地区蒙汉族儿童正常人群HLA-DRB1基因多态性研究

The study on HLA-DRB1 gene polymorphisms in normal juveniles of han people and mongolian residing in inner mongolia

摘要 目的研究内蒙古地区蒙、汉族儿童正常人群HLADRB1等位基因的分布概况。方法采用分析性研究策略,引入PCRSSP技术,对祖籍三代居住内蒙古地区,无血缘关系和与异族通婚史的蒙、汉族正常健康儿童102名和108名进行HLADRB1等位基因的型别分析和比较。结果(1)蒙、汉族HLADRB1各等位基因均被检出;(2)蒙古族HLADRB1基因频率较高的依次是040x(149%)、120x(137%)和150x(103%),而较低的等位基因依次是1001(2%)、160x(2%)和010x(35%);汉族HLADRB1基因频率较高的等位基因依次是0901(15%)、150x(134%)和040x(118%),而较低的等位基因依次是1001(38%)、070x(42%)和160x(42%);(3)蒙古族HLADRB1070x和120x基因频率分别为92%和137%,高于汉族相应的42%和57%(χ2分别是4061和7318,P分别是0044和0007),差异有统计学意义。而汉族DRB10901基因频率为15%,高于蒙古族45%(χ2=12462,P=0.000),差异有非常显著意义;(4)蒙、汉族间遗传距离(D)为0253。结论内蒙古地区蒙、汉族儿童健康人群中HLADRB1等位基因分布不同,但差异不大,符合中华民族是一个统一整体这一事实。 Objective To study the distribution of HLA-DRB1 alleles in the juvenile of han and mongolian residing in inner mongolia.Methods 102 mongolian and 108 han normal juveniles,who have resided in inner mongolia for at least three generations,without kinship and interethnic marriage,were recruited.The polymorphism of HLA-DRB1 alleles was analysed and compared with the PCR-SSP technique.Results (1)HLA-DRB1 alleles in both Han and mongolian were detected.(2)The higher frequenies of HLA-DRB1 gene in mongolian were *040X(14.9%),*120X(13.7%) and *150X(10.3%),and the lower frequencies of HLA-DRB1 gene in mongolian were *1001(2%),*160X(2%) and *010X(3.5%),respectively.The higher frequencies of HLA-DRB1 gene in han were *0901(15%),*150X(13.4%) and *040X(11.8%),but the lower frequencies of HLA-DRB1 gene in han were *1001(3.8%),*070X(4.2%) and *160X (4.2%).(3)The frequencies of *070X and *120X in HLA-DRB1 gene in mongolian were 9.2% and 13.7%,but the frequencies in han were 4.2% and 5.7%.Compared with the frequencies between the mongolia and the han,there were significant differences(χ2=4.061 and 7.318,P=0.044 and 0.007).The frequencies(15%)of HLA-DRB1 *0901X gene in han was higher than that in mongolian(4.5%),there was significant difference(χ2=12.462,P=0.000).(4)The genetic distance(D) between the han and the mongolian was 0.253.Conclusion The distribution of the HLA-DRB1 alleles in normal juveniles of han and mongolian residing in inner mongolia was different,but not sharply,which means that the Chinese may originate from the same nationality.
