摘要
以葡聚寡糖为模型分子,经甲基化,乙酰氯/盐酸裂解,(+)MNB甲酸荧光衍生化及高效液相色谱分析,建立一种新的寡糖链结构分析方法。该方法可以同时获得组成单糖种类、糖链分支位点及组成单糖的D,L构型3种结构信息。操作简单,分析灵敏度为0.2pmol,分析周期在5h以内。
The oligoglucose is taken as a model to establish a new method for the analysis of oligosaccharide structure by methylated, AcCl/HCI degradation, ( +) -2-methyl-2-,beta-naphthyl-1 3 -benzodioxole- 4-carboxylic acid ( ( +) -MNB carboxylic acid) fluorescent derivation and high performance liquid chromatography. The information of type monosaccharide contained, branch point of the oligosaccharide chain and the D,L-configuration of the monosaccharide could be obtained at the same time by this method. The operation is simple and the analytical sensitivity is 0. 2 pmol. The total operation period is within 5 h.
