摘要
目的:探讨应用液相色谱-质谱联用技术鉴定黄芩总黄酮提取物各组分的方法。方法:选用Discovery C_(18)柱,以甲醇-水-甲酸(40:60:0.025)为流动相,经紫外(波长为335 nm)检测后,在电喷雾串联质谱负离子扫描方式下,对HPLC-UV图谱中各色谱峰进行一级和二级质谱分析。结果:经与对照品相比较,鉴定了提取物中4个已知的黄酮类化合物,推断了3个未知黄酮苷类化合物可能的结构。结论:该方法可用于快速分析鉴别天然产物提取物中已知和未知化合物的结构。
Objective:To develop an HPLC-MS/MS method for the identification and structural determination of flavonoids extracted from Scutellaria baicalensis Georg.Methods:The extract was subjected to HPLC, using Discovery C_(18) column,methanol-water-formic acid(40:60:0.025)as mobile phase,and 335nm as ultraviolet detection wavelength.A triple-quadrupole tandem mass spectrometer was applied to verify the LC eluate by Q1 scan and product ion scan spectra, which was operated in the negative ion mode.Result:Four known flavonoids were determined and the possible structures of three unknown flavonoid glycosides were deduced based on the chromatography and MS data.Conclusion:The application of this method could be available and helpful for the rapid discovery and identification of constituents from natural products.
