张力对成骨样细胞PGE_2表达的影响 被引量:5

摘要 目的:探讨不同应变强度的机械张力刺激对体外培养的成骨样细胞PGE2表达的影响。方法:采用四点弯曲细胞力学加载仪对人成骨样细胞MG 63分别以 1 000、2 000、3 000、4 000、6 000、8 000μstrain的力学应变强度进行张力刺激,采用放射免疫法对受不同强度张力作用后成骨样细胞PGE2 的表达分别进行检测并相互比较。结果:成骨样细胞MG 63在受到强度为 1 000μstrain的张力刺激后,其PGE2 的生成量略降低但与对照组无显著性差别,随着张力强度的增加,其PGE2 的生成量显著升高。结论:不同强度的机械张力刺激能够不同程度的增强成骨样细胞PGE2 的表达,PGE2 信号转导途径是将力学刺激信号转入成骨细胞并促进成骨样细胞增殖的重要信号转导途径。 Objective: To investigate the effect of strech on the production of PGE_2 from the osteoblast-like cells.Methods:The osteoblast-like MG-63 cells were stretched for 24 h with the strain of 1 000,2 000,3 000,4 000,6 000 and 8 000 μstrain respectivly by four-point-bend loading device,then the production of PGE_2 from the MG-63 cells was detected by radioimmunoassay. Results:The PGE_2 production(pg/ml) of the control cells was 276.39± 7.77,that of the cells treated by 1 000,2 000,3 000,4 000,6 000 and 8 000 μstrain was 262.07±5.96(vs control,P> 0.05),319.97±11.83,337.20±13.13,424.65±15.53,450.53±14.23 and 508.38±9.26 respectively(each group vs control or vs each of the another stretched group,P<0.05).Conclusion: The stretch load with different amplitude can promote the release of PGE_2 by osteoblast-like cells in a amplitude-depend manner in a certain range.
