脑缺血大鼠干扰素γ与细胞间黏附分子-1表达相关性研究 被引量:2

摘要 目的研究脑缺血时干扰素γ与细胞间黏附分子(ICAM)1的表达是否具有相关性。方法将雄性SD大鼠54只随机分成6组(A、B、C、D、E、F组),每组9只。再将每组大鼠随机分成实验组(6只)和对照组(3只)。A、B、C、D、E、F实验组用埋线法制造大鼠局灶性脑缺血再灌注模型,分别于再灌注后1、3、6、9、12、24h处死大鼠,取大鼠脑组织,应用免疫组织化学方法分别检测干扰素γ及ICAM1的表达。高倍显微镜下计数10个不同视野阳性细胞的平均值。结果大鼠脑缺血再灌注1、3、6、9、12、24h时,干扰素γ及ICAM1的表达逐渐增高,其中干扰素γ阳性的细胞是圆形、以细胞核为主的淋巴细胞,1h阳性细胞表达数为3.2±1.3;6h为7.0±2.2;24h为14.8±1.8。ICAM1阳性细胞是扁平的血管内皮细胞,1h的表达数为12.1±2.1;6h为19.3±3.2;24h为27.6±2.8。结论脑缺血时干扰素γ、ICAM1的表达随时间延长动态地增高,两者具有相关性。干扰素γ也可能是脑缺血时ICAM1表达上调的前炎性细胞因子。 Objective To study assessing th e relationship between IFNγ and ICAM-1 in cerebral ischemia. Methods 54 male Sprague-Dawley rats were subjected to 1 hour of ischemia by ins erting a suture into the lumen of the internal carotid artery and occluding the origin of middle cerebral arterial (MCA), followed by 1~24 hours of reperfusion and recovery. 6 groups (n=9 in each group) of animals were inve stigated after reperfusion of 1, 3, 6, 9, 12, 24 hours separately. Each group ha d a control group (n=3) which underwent a sham operation. Immuno histochemical methods were used to detect the expressions of IFNγ and ICAM-1. Results Both IFNγ and ICAM-1 expression increased at 1, 3, 6, 9, 12, 24 hours after cerebral ischemia and reperfusion. IFNγ positive cells were round nucleus-dominative lymphocyte. The expression of 1 hour was 3.2± 1.3; that of 6 hours was 7.0±2.2; that of 24 hours was 14.8±1.8. ICAM-1 positive cells were flat microvessel endothelial cells of brain. The expression of 1 hour was 12.1±2.1; that of 6 hours was 19.3±3.2; that of 24 hours wa s 27.6±2.8. Correlation analysis indicated the expressions of ICAM-1 and IF Nγ were significantly positively correlated. Conclusion The expressions of IFNγ and ICAM-1 increased and they are correlated after focal cerebral ischemia. It illustrated IFNγ may be a proinflammatory cytokine to enh ance the expression of ICAM-1 in cerebral ischemia.
