摘要 目的探讨缺血前经门静脉注射还原型谷胱甘肽(GSH)对大鼠肝脏缺血再灌注损伤的保护作用。方法20只雄性SD大鼠建立部分肝脏缺血再灌注损伤模型,随机分2组,每组10只:缺血再灌注组(IR组),还原型谷胱甘肽预处理组(GPC组)。测再灌注末血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)、丙二醛(MDA)及GSH含量。结果肝脏缺血45min再灌注40min后,GPC组血清ALT、AST、LDH、MDA含量明显低于IR组(P分别<0.05,0.01,0.01,0.01),而GSH含量明显高于IR组(P<0.01)。结论缺血前经门静脉注射还原型谷胱甘肽预处理可以有效减轻肝脏缺血再灌注损伤程度。其机制可能为增加血循环中GSH,减少肝组织及血循环中自由基含量。 Objective To investigate the protective effect of glutathione(GSH) on the rat liver ischemia reperfusion injury,injected from the stem of potal vein prior hepatic ischemia. Methods 20 male SD rats were randomized into two groups:IR group and glutathione preconditioning group(GPC group). ALT、AST、LDH、MDA and GSH level in serum were measuered. Results After 45 min ischemia and 40 min reperfusion,ALT、AST 、LDH and MDA level in the serum of the GPC group were significantly lower than IR group's(P<0.05,P<0.01,P<0.01,P<(0.01)),but the GSH level was significantly higher than IR group's(P<0.01).Conclusion Injection of GSH from potal vein prior hepatic ischemia can protect hepatic cells form the damage induced by IR injury. The main possible reason were that it might increase the content of GSH and reduce free radical in blood and heptic tissue.
