补阳还五汤对体外低糖低氧损伤海马神经干细胞增殖的影响 被引量:9

The effects of Buyang Huanwu Decoction on the Expansion of Neural Stem Cells Injured by Hypoxia and Glucose Deprivation in Vitro

摘要 目的探讨体外低糖低氧损伤海马神经干细胞(neuralstemcells ,NSCS)后干细胞的反应及补阳还五汤对其的影响。方法从新生wistar大鼠海马分离、培养NSCS,低糖低氧体外模拟脑缺血损伤,随机分组,给予不同处理,运用MTT法、克隆计数观察损伤后NSCS 的增殖情况。结果低糖低氧损伤后部分NSCS死亡,NSCS克隆数为( 65 1 .6±3 5 .3 )个,含药血清加入后克隆增大,克隆数增多,其中治疗1组为( 693 .4±3 3 .3 )个,治疗2组为( 72 4.7±3 1 .4)个,与模型组比较差异有显著性(P <0 .0 5~0 .0 1 )。MTT检测OD值有相似结果,但只有治疗2组与模型组差异有显著性(P <0 .0 5 )。结论补阳还五汤可促进低糖低氧损伤NSCS Objective To explore the expansion of neural stem cells injured by hypoxia and glucose deprivation in vitro and the effects of Buyang Huanwu decoction on it.Methods NSC S were isolated and cultured from neonate rat hippocampus at first,and then ischemic models in vitro were induced by hypoxia and glucose deprivation and divided into 4 groups randomly.The expasion of NSC S was valued by MTT and clone formation.Results Some NSC S died after being injured by hypoxia and glucose deprivation,while the number of neurosphere was 651.6±35.3.Added with herb medicateal serum, both the number and radius of neurosphere were euhanced.The numbers in treating group 1 and in treating group 2 were 693.4±33.3 and 724.7±31.4,remarkably higher than that in modeling group(P<0.05~0.01).Optical density with MTT had similar tendency, but there was significant difference only between modeling group and treatig group 2 (P< 0.05 ).Conclusions Buyang Huanwu decoction can enhance the proliferation of neural stem cells injured by hypoxia and glucose depvivation.
