摘要 腺苷13,130mg·kg-1抑制小鼠的迟发型超敏反应,10,100,1000μmol·L-1抑制小鼠外周全血的淋巴细胞增殖反应,脾淋巴细胞增殖反应和小鼠脾细胞产生白细胞介素2。在人外周血淋巴细胞转化反应中1000μmol·L-1才可见明显的抑制作用;氨茶碱22μmol·L-1逆转1000μmol·L-1的作用,潘生丁10μmol·L-1部分逆转1000μmol·L-1的作用,提示腺苷可能通过腺苷受体而起作用。 Adenosine (Ado) 13,130 mg ·kg-1 ip inhibited murine delayed type hypersensitivity reaction induced by sheep red blood cells;10,100, 1000μmol· L-1 in vitro inhibited PHA induced murine peripheral blood lymphocyte transformation of [3H] TdR incorporation; 1,100 μmol·L-1 in vitro simply enhanced PHA induced human pe-ripheral blood lymphocyte transformation,but 1000 μmol· L-1 obviously suppressed it; 1μmol· L-1 increased Con A (4 and 8 mg· L-1)induced murine spleen T lymphocyte cells mitogensis, but 100,1000 μmol· L-1 inhibited it; 10, 100, 1000 μmol·L-1 obviously inhibited interleukin 2 produced by murine splenocytes(1×109· L-1).Dipyridamole (Dip) 10 mg·kg-1 ip obviously reversed the suppression effects of Ado 130mg· kg-1 ip on murine delayed hypersensitivity reaction,increased effects of Ado 1000μmol·L-1 on murine and human peripheral blood lymphocyte transfor mation, and reversed suppression effects of Ado 1000μmol· L-1 on interleukin 2 produced by murine splenocytes (1×109· L-1). Those results suggested that the uptake of Ado play an important role in cellu-lar immunity. Aminophylline (Ami) 22 μmol·L-1 in vitro could obviously reversed suppression effects of Ado 1000 μmol·L-1 in vitro on PHA induced murine and human peripheral blood lymphocyte transformation of [3H] TdR incorporation,interleukin 2 Produced by murine splenocytes (1×109·L-1).Ami 100 mg·kg-1 ip obviously reversed suppression effects of Ado 130mg· kg-1 ip on murine delayed type hypersensitivity reaction induced by sheep red blood cells. Those results suggested that the suppression effects of Ado on cellular immunity may be mediated by Ado A2 receptor.
