摘要
目的建立口腔鳞癌组织中脂肪酸合酶(FAS)基因表达的相对定量实时PCR(realtimequantitative,PCR)检测,探讨口腔癌中FAS表达的临床意义。方法利用实时定量PCR法测定18例口腔癌患者新鲜癌组织和口腔正常组织标本中FAS表达情况,结合内参照GAPDH基因进行相对定量分析。结果FAS在癌组织中高表达,鳞癌组织中FAS基因表达值为1.42±0.51(FAS/GAPDH),正常口腔组织中FAS基因表达值为1.23±0.50,两组间差异有统计学显著意义(P<0.01)。癌组织分化程度与FAS表达水平没有明显关系。结论实时定量PCR方法能够对口腔鳞癌组织FAS的表达水平进行定量分析,癌组织和口腔正常组织中的FAS表达有显著差异。
Objective To develop a method of relative quantification of fatty acid synthase (FAS) mRNA in squamous cell carcinoma of the oral cavity by using of real-time quantitative PCR.Methods The FAS mRNA expression, including tissues of carcinoma and normal tissues, in 18 cases of oral squamous cell carcinoma were evaluated by real-time quantitative PCR, and quantified according to an internal control GAPDH.Results FAS mRNA was detected in all 36 surgical specimens. The expression of FAS of squamous cell carcinoma was higher than that of normal tissues with a relative copy numbers 1.42±0.51(FAS/GAPDH).FAS mRNA relative copy numbers were 1.22±0.50 in normal tissues. The differentiation degree of squamous cell carcinoma is not relative to the FAS gene expression.Conclusion The FAS mRNA expression can be detected by real-time quantitative PCR technique. There is a notable difference between squamous cell carcinoma and normal tissues of the oral cavity in FAS expression.
