摘要
本研究用化学发光法测定:(1)人血多形核白细胞(PMN)受佛波醇(PMA)刺激发生呼吸暴发产生的活性氧。(2)黄嘌呤-黄嘌呤氧化酶体系产生的超氧阴离子自由基(O2)。(3)维生素C-铜-酵母多糖体系产生的羟自由基(·OH)。(4)过氧化氢(H2O2)释放反应。同时也观察了川芎嗪对上述体系产生的自由基的影响。结果提示,川芎嗪对PMN细胞呼吸暴发的全过程均有抑制作用,对O2有清除作用,而对·OH则无作用。
Chemiluminescence method was used to measure (1) Active oxygen production induced by respiratory burst of polymorphonuclears(PMN) from human blood stimulated with phorbol myristate acetate (PMA); (2)Superoxide(O2) induced by xanthine-xanthine oxidase system, (3)Hydroxyl radicals(OH) produced by Vit C-Cu2+-zymosan; and (4) The release of hydrogen peroxide(H2O2). Effects of tetramethylpyrazine on these active oxygen species were observed. The results showed respiratory burst of PMN was inhibited by tetramethylpyrazine, superoxide and hydrogen peroxide were scavenged by tetramethylpyrazine and their median iuhibition concentration(IC50, μmol . L-1) were 5.6 and 7.1 respectively.
