摘要
在(S,S)WhelkO1手性柱上对去甲羟基安定、扁桃酰胺和氯噻酮3种碱性药物进行了对映体分离研究。考察了流动相正己烷体系中不同种类和浓度的醇类添加剂、不同浓度乙酸对3种碱性药物对映体分离的影响,并初步探讨了其手性分离机理。结果表明,去甲羟基安定和扁桃酰胺在正己烷/异丙醇/乙酸为80∶20∶0.1(V/V)条件下都获得了很好的分离,其最佳分离度分别为2.59和3.63,氯噻酮则未获得分离。
Enantioseparations of oxazepam, mandelic acidamide and chlorthalidone on (S, S)-Whelk-O 1 column were investigated. The influences of the concentration and kind of alcohols, concentration of acetic acid on enantioseparation of three basic drugs were studied. Besides, chiral separation mechanism of these drugs was also discussed. The results showed that oxazepam and mandelic acidamide were separated successfully. In the mobile phase of n-hexane,iso-propanol and acetic acid (80∶20∶0.1, V/V) , the ~resolution of ~oxazepam′s and mandelic acidamiale′s were 2.59 and 3.63, respectively. But chlorthalidone was not got chiral separation.
