摘要
目的:观察中药藤黄的有效成分藤黄酸对胰腺癌细胞是否具有生长抑制和诱导凋亡作用,以挖掘研发治疗胰腺癌的新药物。方法:采用MTT比色法、形态学观察和流式细胞术(FCM)分析研究藤黄酸对于胰腺癌细胞株PC3的作用。结果:倒置显微镜观察发现,藤黄酸作用后的细胞间接触松散,胞浆中颗粒增多,细胞周围出现碎片,部分死细胞漂浮。藤黄酸对胰腺癌细胞株PC3的抑制作用与作用时间有一定的依赖关系,即药物作用时间越长则抑制率越高,而药物浓度与抑制率关系不明显。流式细胞术分析用药后细胞发现凋亡峰,且提示藤黄酸可抑制S期细胞向G2/M期移行。结论:藤黄酸对于胰腺癌细胞具有一定的抑制作用和诱导凋亡作用,应予进一步研究。
Objective:To explore new remedy for pancreatic cancer, the growth condition of pancreatic cancer cell line PC-3 after treated with Gambogic Acid was observed.Methods:MTT and morphology were used to observe the growth suppression, FCM to analyze the apoptosis. Results:With inverse microscope, the following phenomena were observed:cell contact becomes lossing and much more granules appear in cell plasma. There were some debris around cells and some dead cells float in the culture. The cell inhibition after Gambogic Acid treated is time-dependent and not concentration-dependent. Apoptosis peaks were observed and cells arrest from S phase to G_2/M phase after Gambogic Acid. Conclusion:Gambogic Acid may suppress pancreatic cancer cell growth and induce it apoptosis. It is suggested the drug should be further researched.
