摘要
目的:观察甲壳质衍生物—壳寡糖在新西兰兔骨折愈合中的作用及其作用机制。方法:用新西兰兔制备右桡骨中段3mm骨缺损模型后,每日灌胃给予壳寡糖0.28g/kg。术后9、17、30、42d4个时间段,X线片、地衣红染色观察骨折愈合情况;免疫组织化学方法观察转化生长因子β1(TGF-β1)蛋白的表达,并用VIDAS图像分析系统进行半定量分析。结果:实验组在各时间段的骨折愈合情况均显著好于对照组;在9、17d时,实验组骨折端中TGF-β1的表达均较对照组明显增高。结论:壳寡糖能促进兔骨折的愈合,其机制可能与其促进胶原形成、提高骨折愈合早期TGFβ1的表达有关。
Objective: To study the effect of chitooligosaccharide on fracture healing in New Zealand rabbits and its mechanism. Methods: New Zealand rabbits were made into models with 3 mm bone defect in middle segment of radius and were given chitooligosaccharide (0.28 g/kg) daily through stomach tube postoperatively. Four groups were divided according to intervals of 9,17,30 and 42 d postoperation. X-ray and orcein staining were used to observe the healing. Immunohistochemistry was applied to observe the expression of transforming growth factor β1 (TGF-β1) proteins.A semi-quantitation was performed by VIDAS analysis system. Results: Fracture healing of treatment group was better than that of control groups in all intervals. The expression of TGF-β1 in treatment group of 9 and 17 d intervals was significantly higher than that of the control group. Conclusion: Chitooligosaccharide can accelerate fracture healing throgh promoting collagen formation and increasing the expression of TGF-β1 in the fracture healing process.
