摘要 目的 研究bcl -2反义核酸能否增加60 Coγ线对恶性淋巴瘤细胞凋亡的作用。方法 采用细胞染色方法观察细胞凋亡的形态 ,原位末端标记法检测凋亡细胞 ,流式细胞仪检测细胞的DNA含量和bcl 2蛋白的表达。结果  1~ 8Gyγ线和 10~ 4 0 μmol/Lbcl- 2基因反义核酸均能抑制B淋巴瘤细胞株Raji细胞生长 ,诱导细胞凋亡。流式细胞仪检测亚G1期细胞增多 ,bcl- 2蛋白表达降低 ,呈现时间剂量依赖效应 ,联合应用比单独应用抑制作用显著。结论 bcl- 2反义核酸能够增加60 Coγ线对恶性淋巴瘤细胞凋亡的作用。 Objective To investigate whether bcl-2 antisense oligodeoxy nucleotid e (ASODN) could enhance 60Co γ-ray induced-apoptosis in mal ignant lymphoma cells . Methods Cell apoptosis situation, cellular DNA contents and bcl-2 protein exp ression were determined by cell stain, TdT-mediated dUTP Nick-end Labeling (TU NE L) and flow cytometry. Results One to 8?Gy γ-ray and 10-40?μmol/L bcl-2 ASODN could inhibit Raji cell growth and induce cell apoptosis. Assay of flow cytometr y showed that the number of Sub-G 1 cells was significantly increased and Bcl -2 p rotein expression in Raji cells was significantly downregulated, which showed ti me- and dose-dependent manners. The effects were more pronounced when γ-ray and bcl-2 ASODN were used together rather than either alone. Conclusion Bcl-2 antis ense oligodeoxynucleotide could enhance 60Co γ-ray induced -apoptosis in malignant lymphoma cells.
