摘要
目的:研究VC、VE对H2O2诱导的血管内皮细胞氧化损伤的保护作用。方法:采用人脐静脉血管内皮细胞株ECV-304,在培养液中加入不同浓度的VC、VE孵育24h,H2O2诱导损伤,再培养4h后,终止培养。观察细胞形态学变化并检测细胞活力、蛋白质及MDA含量。结果:正常对照组及VC、VE各剂量组细胞活力、蛋白质含量均高于H2O2损伤对照组,差异有显著性(P<0.01);损伤对照组MDA含量高于正常对照组和VC、VE组,差异有显著性(P<0.01)。结论:VC、VE可以减轻H2O2对血管内皮细胞的氧化损伤,具有一定保护作用。
Objective To study the protective effect of vitamin C and E on vascular endothelial cells which were damaged by hydrogen peroxide (H2O2). Methods The oxidative insult model of human umbilical vein endothelial cells (ECV-304) was induced by H2O2. for 4 h. The cells were pretreated with three concentrations of vitamin C and E solution for 24 hours before they were exposed to H2O2. The MTT protein levels in cells and the contents of cellular MDA were determined after 4 h. Results The MTT and protein levels in cells in normal control group and VC or VE treated groups were higher than that in H2O2 damaged group without VC or VE(P<0.05). The level of MDA in H2O2 control group was remarkable higher than that in the other groups(P<0.05). Conclusion Vitamin C and E can prevent the oxidative injury of vascular endothelial cells following hydrogen peroxide.
