摘要
应用14例配对的肝切除标本,系统性地探讨了肝癌和癌旁组织c-myc基因扩增、mRNA转录和c-myc基因产物表达水平的变化,肝癌和癌旁组织未见c-myc基因扩增。原位杂交检测可见11例癌组织,10例癌旁组织mRNA转录水平增加,P62myc免疫组化显示,染色阳性率分别为癌组织85.7%,癌旁组织92.9%。结果表明,人肝癌和癌旁组织存在着转录和基因产物水平高频率的超表达。统计学分析表明两者之间有高度的一致性,结论是c-myc基因超表达与基因扩增无关,不伴有扩增的超表达是肝癌在分子水平呈现的一个重要征象。
The levels of amplification,transcriptions and gene product expression of c-myc gene in tumor tissues and corresponding non-tumor tissues were system atically studied with 14 pairs of resected samples from patients with hepatocellular carcinoma(HCC).The amplification of c-myc gene was not found in tumor tissues and non-tumor tissues using dot blot hybridization technique,Overexpression of c-myc mRNA transcription was detected in 11 tumor tissues and in 10 paired non-tumor tissues using in situ hybridization technique.The positive rate of P62 myc was 85.7%(12/14)in tumor tissues and 92.9%(13/14)in non-tumor tissues respectively.A closely correlation was noticed between c-myc mRNA and gene product expression.The result suggested that amplification was not related to the overexpression.The overexpression of c-myc mRNA and gene product without amplification may be an important sign of HCC at the mollecular level.
