Simplified Enzyme-Linked Immunoadsorbent Assays for Myelin Basic Protein and Antibodies to Myelin Basic Protein

报告一种简易的髓鞘碱性蛋白（ＭＢＰ）及其抗体（Ａｎｔｉ－ＭＢＰ）酶联免疫吸附同步定量测定法。用人脑ＭＢＰ为抗原制备抗血清和辣根过氢化物酶标记兔抗人ＭＢＰ抗体，酶标记物稳定至少２年，ＭＢＰ和Ａｎｔｉ－ＭＢＰ的最低检出量分别为０．２５ｎｇ／ｍｌ和０．２０ｎｇ／ｍｌ，２４小时内即可同时完成两项检测。

Synchronous quantitative determina-tion using simplified enzyme-linked immuno adsorbentassay has been developed for myelin basic protein(MBP)and antibodies to myelin basic protein(Anti-MBP).Antisera were prepared by immunizing rabbitswith purified MBP from human white matter. Anti-bodies to MBP was conjugated with horseradish perox-idase(HRP)by using modified periodate oxidationmethod.Minimum detection amounts of MBP andAnti-MBP are 0.25 ng/ml and 0.20 ng/ml respective-ly.The activities of Anti-MBp-F、P conjugates arestaMe(stored at-30℃) for at least 2 vears. The de-tection procedure can be accomplished within 24hours.
