摘要 目的研究灰树花多糖(Polysaccharideofgrifolafrondosa,PGF)对S180肉瘤的生长抑制作用并探讨PGF抗肿瘤的机制。方法制备荷瘤小鼠动物模型,以PGF溶液和环磷酰胺(cyclophosphane,Cy)溶液分别灌胃10d,检测小鼠的胸腺指数、脾指数、抑瘤率,用免疫组化的方法检测各组瘤块内肿瘤坏死因子-alpha(tumornecrosisfac-tor-alpha,TNF-α)的表达。结果PGF、Cy对S180荷瘤小鼠的抑瘤率分别为47·92%、41·55%,PGF组荷瘤小鼠脾脏指数、胸腺指数明显高于Cy组、生理盐水组,PGF组瘤组织内大量表达TNF-α,Cy组、生理盐水组瘤内不表达TNF-α。结论PGF具有抗肿瘤作用,并且促进荷瘤小鼠大量分泌TNF-α,瘤组织内表达TNF-α可能是灰树花多糖抗肿瘤的方式之一。 Objective: To investigate the effects of Polysaccharide of grifola frondosa on the growth of S-180 sarcoma, and further demonstrate its mechanisms. Methods: S-180 sarcoma cells were transplanted into mouse subcutaneous tissue to obtain the animal model, and polysaccharide and cyclophosha solution was injected into mouse stomach for 10 days, and thymus Index, spleen Index, tumor inhibitory rate were evaluated. The imanunohistochemistry methods were used to measure the expression levels of TNF-α. Results: Inhibitory rate of S-180 sarcoma was 47.92% and 41.55% after the administration of Polysaccharide and cyclophosphane respectively. Thymus Index, spleen Index in the polysaccharide group were markedly higher than those in the cyclophosphane and Saline groups. The expression level of TNF-α in polysaccharide group was significantly higher than that of the cyclophosphane and Saline groups. Conclusion: Polysaccharide of grifola frondosa can inhibit tumor growth and promote secretion of TNF-αin inhibitory of S-180 sarcoma mass which may be one of mechanisms of Polysaccharide of grifola frondosa.
