摘要 目的探讨雷公藤内酯醇对血清诱导的大鼠血管平滑肌细胞(VSMCs)增殖的影响及其作用机制。方法体外培养大鼠胸主动脉VSMCs,细胞融合达60%时培养基中加入不同浓度的雷公藤内酯醇和雷帕霉素。采用细胞计数法观察细胞增殖情况,流式细胞仪分析细胞周期;RTPCR法检测VSMCs内cfosmRNA表达水平。结果雷公藤内酯醇和雷帕霉素均明显抑制血清诱导的大鼠VSMCs增殖,阻断细胞由G0/G1期向S期转化;RTPCR检测显示,随着雷公藤内酯醇浓度的增加,cfosmRNA表达明显减少。结论雷公藤内酯醇能明显抑制血清诱导的VSMCs增殖,而阻断细胞周期、下调原癌基因cfosmRNA的表达可能是其药理机制之一。 Objective To explore the effects and mechanism of Triptolide on the proliferation of cultured vascular smooth muscle cells (VSMCs). Methods Aortic smooth muscle cells of rats were cultured and characterized in vitro. Using rapamycin as positive control, we used cell counting to investigate the effects of Triptolide and rapamycin on the VSMCs. Flow cytometry was used to study the influence of Triptolide and rapamycin on the VSMCs cell cycle. The effects of Triptolide on c-fos mRNA expression were measured by RT-PCR. Results Triptolide and rapamycin inhibited proliferation of VSMCs induced by blocking cell cycle progress from G0/G1 phase to S phase. It was founded that Triptolide inhibited c-fos mRNA expression in a concentration dependent maner. Conclusion Down-regulation of proto-oncogen c-fos mRNA expression by Triptolide may be one of the mechanisms with which Triptolide displays its inhibitory effect on VSMCs.
