摘要
利用正交试验方法研究了培养基中主要成分,NH,NO,Ca ̄(2+),Mg ̄(2+)对党参体细胞胚胎发生的影响。发现30mmol/LNH+30mmol/LNO组合最有利于体细胞胚胎发生,其它依次为30mmol/LNH、30mmol/LNO、30mmol/L NH与0.5mmol/LCa ̄(2+)、30mmol/LNO与7.0mmol/LCa ̄(2+)、3.0mmol/LCa ̄(2+)、1.5mmol/LMg ̄(2+)。通过分析得出优选培养基成分为30mmol/LNH、30mmol/LNO、1.0mmol/LCa ̄(2+)和1.5mmol/LMg ̄(2+),其它成分均同MS培养基成分。用此培养基,党参体细胞胚胎发生比MS培养基提高2.5倍。
The main composition of culture medium such as NH,NO,Ca ̄(2+),Mg ̄(2+) were tested fortheir effects on Dangshen [Condonopsis pilosula (Franch)Nannf.] somatic embryogenesis fre-quency by means of Orthogonal experiment. The result showed that the interaction of 30 mmol/L NH and 30 mmol/L NO played an important role in induction of embryogenesis. The otherin order: 30 mmol/L NH, 30 mmol/L NO_3, 30 mmol/L NH and 0. 5 mmol/L Ca ̄(2+),1.5mmol/L Mg ̄(2+), Analysing the result and getting this medium compositions are as follows : 30mmol/L NH.30 mmol/L NO_3 ,1. 0 mmol/L Ca ̄(2+),1. 5 mmol/L Mg ̄(2+),others are the same asMS medium.This medium increase somatic embryo yield 2. 5 fold.
