摘要 采用四唑盐比色法(MTT)、3H-TdR掺入试验、细胞大小测定的图象分析等方法,观察不同浓度牛血浆纤维粘连蛋白(FM)和人工合成肽段(RGDS,40μg/ml)对体外培养的人牙髓细胞的影响。FN(20、40μg/ml)可促进牙髓成纤维细胞增殖、刺激细胞DNA合成,FN(10、80μg/ml)、RGDS(40μg/ml)和空白组对牙髓细胞无上述效应。4种浓度FN和RGDS(40μg/ml)均使牙髓细胞处于伸展状态,支持细胞的粘附。说明FN可促进牙髓细胞增殖,并可通过整合素受体调节细胞与基质之间的联系。 The effects of fibronectin(FN)on cultured human dental pulp cells in vitro were studied.Four concentrations of bovine plasma FN(10,20,40,80μg/ml),one concentration of Arg-gly-asp-ser peptide(RGDS,40μg/ml) were added to the media.Control cultures were made in the absence of FN and RGDS.FN(20,40μg/ml) stimulated the cells proliferation using MTT colorimetric assay and promoted incorporation of[3H]thymidine.FN(10,80μg/ml),RGDS(40μg/ml)and control experiment had no such abilities.FN and RGDS also kept the pulp eclls in spreading state.The results suggest that FN is involved in the regulation of pulp eclls proliferation,and the cell-matrix interactions are partly mediated by receptors related to the integrin family.
