慢粒白血病细胞的分化潜能及TPA的诱导作用

摘要 慢粒白血病细胞在体外液体培养条件下具有一定的分化成熟能力。表现在细胞数目增多,NBT还原能力增强,CE强阳性细胞比例增高,杆状和分叶核粒细胞增多.TPA(1.6×10^(-7)M)则诱导此类细胞向单核巨噬细胞分化。细胞由悬浮转为贴壁,胞浆丝状伪足突出并相互连接形成网状结构,形态与细胞化学反应具有单核巨噬细胞特征。此项实验对于了解慢粒白血病细胞的增殖分化潜能及TPA的作用特点有一定意义。 The differentiative potential of leukemic cells from 8 patients with chronic myelogenous leukemia (CML) in chronic phase and the effect of phorbol diester (TPA) on these cells were observed. In the absent of TPA, most of the immature myeloid cells could undergo spontaneous granulocyte differentiation in liquid culture. The number of total viable cells increased to about 207.2% of the initial values on day 6 and the band and segmented neutrophils increased. Also, the activity of reduced nitroblue tetrazolium (NBT) and chloroacetate esterase enhanced significantly (P<0.001). In contrast, TPA (1.6× 10^(-7)M) induced macrophage-like differentiation of CML cells. The cells became adherent to the culture dishs promptly, extended long pseudopodia, formed cell cell connection and showed remarkable changes of macrophage morphology and cytochemical stain. These data may be of value in understanding the differentiation of CML cells and the effect of TPA.
