摘要
以黑腹果蝇OregonK品系列评价了云南地方中草药昆明山海棠抽提物(THH)在动物生殖细胞中的非整倍体诱发效应,秋水仙素(COL)为本试验的阳性对照物。结果发现,在所有雌雄成虫口饲染毒受试组中,THH(10.80mg/ml)及COL(2.5.20ug/ml)均显著诱发生殖细胞非整倍体,从而导致XO及XXY例外子代频率显著升高(p<0.001.0.05).试验证明:THH及COL均能够在果蝇生殖细胞形成过程中,导致染色体的丢失或不分离;雄性果蝇对受试物诱发生殖细胞X染色体遗失效应较为敏感,而雌性果蝇则易检出诱发生殖细胞性染色体不分离的化合物。
The Oregon K system of Drosophila melanogaster Was emploied for the detection of germline aneuploidy induced by Tripterygium Hypoglaucum(Lev1)Hutch(THH)and Colchicine (COL).Adult flics were treatcd by feeding the compounds.THH(10~80mg/ml)and COL(2.5~20ug/ml)signinca- ntly induced the exceptional offspring dericed from aneuploid mategermcells,but induccd low of those (still signiflcant)derived from treatcdparcntal femalc The cxperiment indicated that THH and COL arcancuploi-dogens in Drosophila germlinc.Mate ftics may be moresensitive to the chemi-cals which induce chromosomc loss,while thefemales mav use to detect the chemicals which share the potency of nondisjunction induction.
