犬肺巨噬细胞的制备及其对不同毒力犬瘟热病毒的敏感性

Preparation of Dog Lung Macrophage and It's Susceptivity to Infection with Virulent and Attrnuated CDV

犬肺巨噬细胞（ＤＬＭ）是犬瘟热病毒允许性细胞系统．本文报道了制备高纯度、高活性ＤＬＭ的方法．通过本方法制备的巨噬细胞培养物纯度达９９％以上，活细胞数占９１％．１只２～６周龄小犬的肺可制备５×６７～９×１０７细胞．病毒的敏感性试验表明，ＤＬＭ不但能支持犬温热强毒增殖，而且还能支持犬瘟热弱毒的增殖且其感染力无明显差异．因此可以得出结论：犬瘟热病毒对大和水貂的毒力并不总是与该病毒在ＤＬＭ上的感染和复制能力相一致的。

Abstract Dog Lung Macrophage(DLM) is a permissive cell system of canine distemper virus. This report describes a method of preparing DLM with high purity and high viability. The purity of macrophages prepared by this method is higher than 99% and 91 % are viable,A lung of puppy 2 to 6weeks of age can yield approximately5×107to 9 ×107cells. The result of CDV infection in DLM shows that all the four virulent and attemuated CDV used in this test can propagate in DLM. We can conelude that CDV virulence was not always linked to its ability to infect or replicate in DLM.
