摘要
小鼠ip和iv胡蔓藤碱甲(Hum)的LD50分别为0.21(95%可信限为0.20-0.22)和0,128(95%可信限为0.119-0.137)mg·kg ̄(-1)小鼠死于呼吸衰竭.大鼠iv和ipHum的LD50分别为0.157(95%可信限为0.149-0.165)和0.26(95%可信限为0.25-027)mg·kg ̄(-1),大鼠也死于呼吸衰竭。大鼠ipHum0.4mg·kg ̄(-1),呼吸停止时,EEG基本平坦成一等电位线,约15min后,ECG才停止。Hum(0.1mg·kgiv,0.01mgicv或0.008mg闩部局部贴敷)可致大鼠隔神经放电节律减慢,放电脉冲数及频率增加,呼气时程明显延长,而吸气时程无明显变化。印防己毒素可明显降低小鼠Hum中毒的死亡率以及延长大鼠Hum中毒的死亡时间。而戊四氮仅能延长大鼠Hum中毒的死亡时间。
Humantenmine(Hum)is a newalkaloid isolated from Gelsemium elegan Banth in China, The LD_(50) of Hum in mice, dead fromrespiratory failure, were 0.21 mg·kg ̄(-1) ip [95% confi-dence limits (CL) were 0.20 to 0.22 mg·kg ̄(-1) and0.128mg·kg ̄(-1)iv(95% CL were0.119 to0.137mg·kg ̄(-1)). The LD50 of Hum in rats,also dead fromrespiratory failure, were 0.26 mg·kg(-1) ip(95% CLwere 0.25 to 0.27 mg·kg(-1)) and 0.157 mg·kg(-1)iv(95% CL were 0.149 to 0.165 mg·kg(-1)), After Hum0.4mg·kg(-1))ip in rats,the respiratory action stopped,at the same time, the voltage of electroencephalogramwas depressed and flatted, but the electrocardiogramstill lasted for about 15 min. Hum 0.1 mg·kg(-1) iv,0.01 mg icv, or 0.008 mg applied onto rat obex de-creased the rate of phrenic nerve discharges(PND), in-creased the frequency of PND,the number of spike inPND,and the time of expiration, while the time of in-spiration was not significantly changed.Picrotoxin de-creased the rate of mortality in mice induced by Humand prolonged the survival duration in rats,butpentylenetetrazol only prolonged the survival durationin rats. The results suggest that Hum maY have directtoxic action on respiratory center and picrotoxin maybe as a potential emergency drug in the therapy oftoxication induced by Hum.
