目的:建立清醒状态下电针治疗全身炎症反应综合征(SIRS)大鼠模型.方法:雄性SD大鼠50只随机分为3组:①内毒素(LPS)组20只;②LPS+电针组20只;③对照组10只.其中组①和组②又分为1.5 h和6 h两个亚组,每个亚组10只.于尾静脉注射LPS(5 mg/kg)制作SIRS模型,对照组注射等量生理盐水.于电针前后检测体温、外周血白细胞数目和血浆肿瘤坏死因子(TNF-α)含量.结果:大鼠电针前体温(T)为(38.73±0.28)℃,白细胞(WBC)为(1.67±1.09)×109/L,TNF-α为(926.97±566.54)ng/L;电针后T为(37.35±0.68)℃,WBC(3.68±0.88)×109/L,TNF-α为(444.66±244.16)ng/L.该剂量引起的WBC数量及体温的变化符合动物SIRS的诊断标准,电针降低了SIRS大鼠血浆中升高的TNF-α水平(P<0.05)并增加其外周血中降低的WBC数量(P<0.01).结论:该模型成功复制了SIRS的病理过程,电针治疗可在清醒状态下减少SIRS大鼠的血浆TNF-α含量,增加白细胞数量.
Objective:To make an animal model in treatment of systemic inflammatory response syndromel SIRS) with electroacupunture (EA) in conscious rats.Methods:A total of 50 rats were divided randomly into three groups:lipopolysaccharide (LPS,n=20),LPS+EA (n=20) and normal control (n=10).Animals of LPS and LPS+EA groups were divided into 1.5 h and 6 h sub-groups respectively (each n=10).LPS (5mg/kg) was injected into the caudal vein in experimental rats,and equal quantity of saline solution was injected into the control animals.Electroacupuncture was performed with 2-100 Hz,2 mA for 1.5 h.The body temperature,leukocyte count of peripheral blood and tumor necrosis factor-α(TNF-α) content in plasma were determined before and after EA.Results:Before electroaupuncture,T:(36.86±0.53)℃,WBC×(1.67±1.09)×10~9/L,TNF-α(926.97±566.54)ng/L;after electroacupuncture,T(℃):(37.35±0.68)℃,WBC×(3.68±0.88)×10~9/L,TNF(444.66±244.16)ng/L LPS injection resulted in significant lowering of leucocyte count and body temperature in experimen- tal rats,and these changes met the criteria of SIRS in animal.Electroacupuncture treatment lowered the content of TNF-αand increased WBC count in animals with endotoxemia(P<0.05,P<0.01,respectively).Conclusion:This rat model was successful in replicating SIRS process.Electroacupuncture therapy could lower the content of TNF-αin plasma and increase WBC count in SIRS rats.
