摘要
目的研究红花黄色素在大鼠各肠段的吸收动力学特征.方法采用大鼠在体肠段灌流实验,主要从药物浓度、pH值、吸收部位等3方面对红花黄色素的肠段吸收特性进行研究.结果在5~20μg·ml-1浓度范围内药物浓度对吸收速率常数无影响;在pH7.8~5.4内药物吸收不受pH值影响;各肠段的吸收速率常数按空肠、十二指肠、回肠、结肠顺序依次下降,分别为0.111 6、0.075 3、0.073 9、0.018 5h-1.结论药物吸收符合一级动力学特征,吸收机制为被动扩散.
Aim To investigate the absorption characteristics and mechanism of safflow yellow for various intestinal segment. Methods The intestine in rats was cannulated for in situ recirculation. The effects of drug concentration, pH value and absorption site on safflow yellow absorption were studied. Results The concentrations of safflow yellow from 5μg·ml^-1 to 20μg·ml^-1 and the pH of drug solution from pH 7.8 to pH15.4 had no distinctive effect on the absorption kinetics. The absorption rate constants (Ka) were 0. 1116,0.0753,0.0739,0.0185h^-1 at jejunum,duodenum,ileum and colon,respectively. Conclusion The absorption of safflow yellow was a first - order process with the passive diffusion mechanism.
