摘要
目的:探讨1,25-(OH)2Vit D3对小鼠牙囊细胞向成骨细胞(成牙骨质细胞)分化能力的影响.方法:第3代小鼠牙囊细胞与10-8mol/L的1,25-(OH)2Vit D3共孵育3、5、7、9 d后,测定其对牙囊细胞增殖、碱性磷酸酶(alkaline phosphotase,ALP)活性、蛋白含量及骨钙素(osteocalcin,OC)表达的影响.结果:在5~7 d的时间范围内,1,25-(OH)2Vit D3能显著促进牙囊细胞的增殖(P<0.05),在5~9 d内能提高ALP活性(P<0.01),增加OC阳性细胞率(P<0.05),但在9 d时能使总蛋白含量下降(P<0.05).结论:1,25-(OH)2Vit D3能促进牙囊细胞的增殖及向成骨细胞方向分化,但对蛋白合成有抑制作用.
AIM :Tu explore the effect of 1,25 - (OH) 2 Vit D3 on cytodifferentiation of mouse dental follicle cells. METHODS:TIle third 3 passage cells were incubated with 10^-8mol/L 1,25 - (OH)2Vit D3 for 3,5,7,9 days respectively. The cell proliferation, amount of total protein, alkaline phosphatase (ALP) , and expression of osteocalcin (OC) were measured. RESULTS:Within the time from 5 to 7 days, 1,25 -(OH) 2Vit D3 promoted dental follicle cells proliferation significantly. It also improved ALP activity( P 〈 0.01 ) and percentage of OC positive cells( P 〈 0.05 ) from 5 to 9 days. However, 1,25 - ( OH ) 2Vit D3 inhibited the amount of total protein content at 9 d( P 〈0.05 ). CONCLUSION:Proper concentration of 1,25 - (OH)2Vit D3 could promote dental follicle proliferation and differentiation towards osteoblasts(cementoblasts) ,trot inhibit protein synthesis.
