摘要
利用双微电极电压钳制技术研究丹参注射液(以下简称丹参)对离体家兔浦肯野纤维慢内向电流(Isi)的电生理作用。结果表明。丹参在浓度为1.0mg/ml 和2.0mg/ml,灌流20 min 时,能明显抑制 Isi,由给药前的76.25±20.83nA 降至53.13±18.70nA 和36.00±10.84 nA(P<0.05和0.01),对Isi 峰值抑制率分别为31%和53%,呈浓度依赖性抑制作用;在浓度为0.5mg/ml,刺激频率为1.0Hz,随着刺激教增加,丹参在开始抑制 Isi 的峰值不明显,以后抑制逐渐加强,直至稳态,呈使用依赖性阻滞作用。以上表明丹参具有钙拮抗剂作用。
Two-microelectrode voltage clamp technique was used to study the effects of Salvia milti- orrhiza on slow inward currents(Isi)of isolated rabbit cardioc Purkinje fibres.The results indi- cated that Salvia miltiorrhiza at 1.0mg/ml to 2.0 mg/ml inhibited Isi respectively from 76.25 ±20.83 nA to 53.13±18.70 nA,and from 76.25±20.83 nA to 36.00±10.84 nA(P<0.05 and 0.01)20 minutes later.Salvia miltiorrhiza inhibited amplitude of Isi respectively about 31% and 53%.It was showed that concentration-dependence blocked Isi.At 0.5mg/ml and a frequency of 1.0 Hz Salvia miltiorrhiza inhibited Isi uncertainly at the beginning.By addition of number of pulses Salvia miltiorrhiza inhibited evidently Isi later,until a steady state oc- curred.It was showed that use-dependence blocked Isi.It was concluded that Salvia miltior- rhiza had function of calcium channel antagonist.
