摘要
优化了中药葛根异黄酮类成分加压溶剂提取的条件。采用高效液相色谱法同时测定葛根提取液中葛根素、大豆苷和大豆苷元含量,以三者含量总和为评价指标,以用正交设计的考察方式对影响加压溶剂提取葛根异黄酮类成分的因数进行优化。优选出最佳提取条件:溶剂为甲醇;温度140℃;提取时间10 m in;压力8.27 MPa,置换体积为60%,并与葛根药典提取方法进行比较,结果表明:加压溶剂提取法具有提取时间短、溶剂消耗少、提取效率高、重现性好、操作模式多样化以及操作过程自动化等优点,将有助于中药质量控制提取过程的标准化。
Isoflavonoids in Pueraria lobata were exdracted by pressurized solvent extraction (PSE). Orthogonal design was used to examine the factor that affects the efficiency of PSE extraction, and the quantitative marker was the total content of Puerarin, Daidzin and Daizein which were determined simultaneously by high performance liquid chromatography. The optimum conditions of PSE extraction of isoflavonoids from Pueraria lobata were as follows: methanol as solvent, temperature at 140℃, pressure under 8.27 MPa, 10 min duration and transpositional volume as 60%. Furthermore, the yields of isoflavonoids from Pueraria lobata by using PSE and extraction method according to the Chinese Pharmacopoeia (ChP) monography was compared. PSE technology has the advantages of shortening the extraction time, decreasing the consumption of solvent, increasing extraction efficiency and automatic operating capabilities.
