摘要
目的初步探讨草麻黄中补体抑制成分(comp lem ent inh ib iting component,C IC)对异种心脏移植超急性排斥反应的作用。方法按是否给予C IC灌胃处理,将动物模型分为对照组和实验组,观察对照组和实验组术后供心存活时间,在术后早期及供心失活时取两组心脏组织行光镜、电镜病理检查,并在术后固定时间分别抽鼠血行血清学检查,用微量法测定CH50、APH50,用透射比浊法测定C3。结果实验组移植心脏存活时间为(51.00±3.46)m in,与对照组(30.30±3.46)m in比较差异极显著(P<0.01)。结论豚鼠至大鼠异种心脏移植后,补体的经典途径和替代途径均被激活。C IC能明显抑制补体活性,延缓超急性排斥反应的发生,从而使移植心脏的存活时间延长。
Objective To study Ephedra sinica in preventing hyperacute rejection in guinea pig-to-rat cardiac xenotransplantation. Methods The recipient rats were divided into two groups: control group and experiment group, according to whether administrated complement inhibiting component (CIC) from Ephedra sinica. In 15 min after operation, 4 rats from control group and 4 from experiment group were sacrificed to detect the pathologic changes in cardiac tissues. The complement parameters including CH50, APH50, and C3 were determined during the period of 0, 10, 15, 30, 45 min in serial serum. Results The survival time was significant- ly prolonged and pathological changes were less in experiment group than that in control group. Conclusion The study reveals that both classical pathway and alternative pathway of complement activation can be induced by xenotransplantation and Ephedra sinica can prolong the xenograft time by inhibiting complement activities and postposing hyperacute reiection.
