摘要 目的:评价重量法在大鼠在体肠单向灌流技术中的应用。方法:以长春西汀为模型药物,采用大鼠在体肠单向灌流技术,对重量法和酚红法计算的净水流量值(NWF)、药物吸收速率常数(Ka)和药物表观吸收系数(Papp)进行比较,并对重量法中收集液在密度修正前后的NWF和Papp值进行比较。结果:重量法计算所得的NWF值显著高于酚红法(P<0.05),前者计算所得的Ka和Papp值略高于后者(P>0.05);重量法能显著减少实验误差;对收集液密度进行修正可以进一步提高实验数据的准确性。结论:重量法可以作为大鼠单向灌流在体肠吸收实验中校正灌流液体积的有效方法,它避免了加入“标示物”给实验带来的一系列问题。 Objective: To evaluate if the gravimetry improves the efficiency of the rat single-pass intestinal perfusion technique. Methods: Vinpocetine was used as a model drug. The values of net water flux (NWF), drug absorption rate constant( Ka ) and drug apparent intestinal permeability( Popp ) by the gravimetry were compared with the ones by phenol red assay. The values of MWF and Papp prior to and post the density correction were analyzed in the gravimetry based on the assumption of a perfusion density of 1.0 g· mL^-1. Results: The values of NWF by the gravimetry were significantly larger than the ones by the phenol red assay ( P 〈 0.05) ; the Ka and Papp by the gravimetry were slightly higher than the ones by the phenol red assay (P 〉 0.05). The gravimetry significantly reduced the systematic errors, and the density correction in the gravimetry provided more reliable results. Conclusion: The gravimetry may significantly correct the perfusion volume in the rat single-pass intestinal perfusion technique, which prevents the results from the systematic errors due to the addition of an external “nonabsorbed” marker.
