摘要
利用随机扩增多态DNA技术对大鸨(Otis tarda)6个个体进行了随机扩增多态DNA分析,共筛选出有效随机引物14条,利用所得的随机引物对每只大鸨的基因组DNA进行扩增,共得到327个扩增片段,其中共有片段150个,根据聚类分析所得到的树状图确定了6只大鸨的亲缘关系.
Randomly amplified polymorphic DNA (RAPD) was used to investigate genetic relationships of 6 Otis tarda . 14 arbitrary primers were screened out and used to amplify genomic DNA of each Bustard. 14 primers had clear and distinct RAPD patterns in each amplification. 327 amplified fragments were generated by RAPD and 150 fragments were shared by the 6 Great Bustards. Hierarchical cluster analysis of the RAPD data was used to calculate the similarity coefficient matrix and dendrogram was constructed. The dendrogram indicates the parent relationships of the 6 Great Bustards.
