鸡主要组织相容性复合体I(BF2和β_2m)二级结构与同源模建 被引量:1

摘要 为了推进鸡主要组织相容性复合体(MHC)Ⅰ类分子(BF2,β_2m)空间结构的研究,克隆、表达了鸡BF2和β_2m基因,采用圆二色谱(CD)测定了其重组蛋白的二级结构,并同源模建了其三级结构。首先,将BF2和β_2m基因分别插入原核表达载体进行了可溶性表达。对表达的融合蛋白进行纯化、蛋白酶切割并对切割后的蛋白进行分离和纯化,获得了麦芽糖结合蛋白(MBP)—BF2, β_2m蛋白以及MBP单体。随后,测定了BF2和β_2m的CD值。CD表明BF2蛋白呈典型的α螺旋; 其中,α螺旋、β折叠、转角和随机蜷曲分别为72,102,70和90个氨基酸。β_2m重组蛋白呈典型的β折叠,α螺旋、β折叠、转角和随机蜷曲分别为0,46,30和22个氨基酸。同源模建显示鸡BF2和β_2m蛋白与人HLA-A2和小鼠H2的3D结构类似。结果显示了BF2和β_2m蛋白在二维和三维上的分子特征及其与抗原多肽结合氨基酸的空间分布,为以后进一步构建鸡BF2-β_2m复合物体外鉴定病毒抗原肽系统奠定了基础。
