摘要
目的构建并评价垂体生长激素腺瘤靶向性基因治疗系统。方法构建GE7系统介导的生长激素启动子调控的基因治疗系统,通过垂体生长激素腺瘤GH3细胞及对照细胞的体外基因转染实验,观察此基因治疗系统对GH3细胞的转染能力和靶向性。结果成功构建基因治疗系统,基因转染后GH3细胞特异性表达报告基因,对照的HO8910PM和U-2OS细胞报告基因的表达不明显。结论应用GE7转染的生长激素启动子调控的基因治疗系统能靶向性地将目的基因转入垂体生长激素腺瘤细胞。
Objective To construct and study the targeted gene therapy system for growth hormone- secreting pituitary tumors. Methods The GE7 system-mediated human growth hormone promotor controlled gene therapy system was constructed. Growth hormone-secreting pituitary tumors cell lines-GH3 cells and control cells were treated with the gene therapy svstem. The targeting and transfection of the therapy system were observed. Results The double targeted gene therapy svstem for growth hormone-seereting pituitary tumors was composed successfully. Reporter gene expressed specifically in GH3 cells. The obvious expression in the HO8910PM and U- 2OS cells were not observed. Conclusion GE7 system-mediated hGHp controlled gene therapy system ean transfer target gene to growth hormone-secreting pituitary tumors effectively and specifieally.
