醒脑启智胶囊药物血清对缺氧诱导的PC12细胞凋亡及凋亡基因的影响 被引量:1

摘要 目的探讨醒脑启智(XNQZ)胶囊药物血清对缺氧诱导的PC12细胞凋亡及其相关基因bcl-2、bax表达的影响。方法采用血清药理学方法,体外培养PC12细胞,以药物血清孵化后用连二亚硫酸钠(Na2S2O4)缺氧诱导,观察PC12细胞凋亡及bcl-2、bax表达的变化。结果缺氧诱导后,PC12细胞的bax蛋白表达上调,使细胞凋亡趋势增强。XNQZ药物血清可使bcl-2基因表达量增加,bax基因表达量降低,细胞凋亡率下降。结论XNQZ可通过调节bcl-2、bax表达,抑制细胞凋亡,提高细胞抗损伤和修复能力,以达到对抗神经元迟发性死亡,抑制智力减退的作用,并呈现一定的剂量依赖关系。 Objective To study the effect of drug serum with Xingnaoqizhi(XNQZ) capsule on PC12 cells apoptosis and apoptosis gene due to damage of hypoxia. Methods PC12 cells cultivated in vitro with method of serum pharmacology were damaged by hypoxia with sodium hydrosulfite,to observe the changes of PC12 cell apoptosis and expressions of apoptosis related gene bcl-1 and bax. Results After damage of hypoxia,bax protein expression of PC12 cell was upregulated,increasing trend of apoptosis. Drug serum with XNQZ capsule could increase bcl-2 protein expression and decrease bax protein expression, reducing apoptotic rate. Conclusions XNQZ capsule could inhibit cellular apoptosis, and increase damage resistance and self-repairment ability of cell by regulation of bcl-2, bax gene expression, which could evidently resist delayed neuronal death and alleviate intellect letdown with the certain dose-dependent relation.
