摘要 目的:探讨复方中药制剂补阳还五汤的抗氧化机制。方法:用水煎醇沉法制备补阳还五汤提取液,从新生鼠坐骨神经分离纯化雪旺细胞,建立过氧化氢(H2O2)损伤模型。将培养细胞分为补阳还五汤处理组、氧化损伤组及正常组和空白对照组,培养12 h时,检测丙二醛(MDA)的含量,用免疫组化方法检测Caspase-3的表达,用RT-PCR技术检测胶质细胞源性神经营养因子(GDNF)的表达。结果:与氧化损伤组相比,补阳还五汤处理组的MDA含量明显减弱,Caspase-3呈弱阳性表达,GDNF mRNA的表达量减少不明显。结论:补阳还五汤具有良好的抗过氧化损伤效果,对H2O2引起雪旺细胞的损伤具有保护作用。 Objective: To explore the antioxidant mechanism of Buyanghuanwutang (BYHWT). Methods:Schwarm cells were purified from the sciatic nerve of neonatal rats and were divided into 3 groups: H2O2 group, BYHWT group and normal group. Malondialdehyde (MDA) content in Schwann cells was determined by biochemical analyzer after 12 h, the expression of Caspase-3 was detected by immunohistochemistry, and the expression of GDNF rnRNA was detected by RT-PCR. Resuits: Compared with H2O2 group, intracellular MDA content in BYHWT group was significantly lower and expression of GDNF mRNA kept at a higher level (P〈0.05). Conclusion: BYHWT has a good antioxidant effect and can be used to protect the Schwarm cell from damage caused by H2O2.
