摘要
目的:检测并鉴定感染组织中的克里米亚-刚果出血热病毒。方法:分别经皮下与腹腔两种途径感染乳小鼠;应用RT-PCR技术对病毒核酸进行检测;利用透射电镜超薄切片技术对病毒进行形态学鉴定。结果:RT-PCR可在心脏、肝脏、脾脏、肾脏以及脑组织内检测到病毒核酸,透射电镜下可在肝细胞胞浆内观察到形态典型的病毒粒子。结论:应用RT-PCR技术可以对感染组织中的克里米亚-刚果出血热病毒进行检测,利用透射电镜可以对病毒的形态进行观察与鉴定。
Objective: To detect and identify Crimean-Congo hemorrhagic fever (CCHF) virus in infected tissues. Methods:The suckling mice were infected with CCHF virus by subcutaneous or intraperitoneal injection, and CCHF virus in the infected tissues was examined by RT-PCR. Morphology of CCHF virus was identified by transmission electron microscopy. Results: The nucleic acid of CCHF virus could be detected in the hearts, livers, spleens, kidneys and brains of the infected suckling mice by the RT-PCR. Virus with typical configuration could be found in the cytoplasm of the hepatocytes under the transmission electron microscope. Conclusion: The virus in infected tissues can be detected by RT-PCR and morphology of CCHF virus can be identified by transmission electron microscopy.
