丹参酮ⅡA对兔脑缺血再灌注损伤的保护作用 被引量:17

摘要 目的研究丹参提纯物丹参酮ⅡA对脑缺血后血脑脊液屏障及细胞内钙的影响。方法24只兔随机分成3组,每组8只,即假手术组(Ⅰ组)、缺血组(Ⅱ组)、治疗组(Ⅲ组)。采用双颈动脉夹闭低血压脑缺血模型,夹闭双侧颈总动脉30min,然后松开血管夹,经颈外静脉回输血液,待血压恢复到术前水平时,静脉注射伊文氏兰(Even’sblue,EB)3mg/kg,Ⅰ组、Ⅱ组经腹腔注射生理盐水3mL,Ⅲ组腹腔注射丹参酮ⅡA(10mg/kg,约3mL),3组均在4h后开颅,取额顶皮层脑组织约100mg,测量脑组织所含EB含量、脑组织含水量(WR)及检测细胞内游离钙浓度([Ca2+]i)。结果Ⅱ组、Ⅲ组EB、WR、[Ca2+]i较Ⅰ组高(P<0.05),血脑脊液屏障通透性增加,缺血区脑组织细胞损伤明显;但Ⅲ组EB、WR均较Ⅱ组低(P<0.05),[Ca2+]i亦低于Ⅱ组(P<0.05)。结论丹参酮ⅡA抑制了血脑脊液屏障通透性的增加,抑制缺血区脑组织细胞内钙超载。可以减少脑组织含水量,一定程度修复血脑脊液屏障。 Objective To evaluate the neuroprotection effect on rabbit forebrain ischemia model treated with Tanshinone Ⅱ A(TS N) by determining cytosolic Ca^2+ concentration. Methods The rabbits were randomly divided into three groups: Sham group (Group Ⅰ , n = 8), Control group (Group Ⅱ , n = 8)and Treatment group (Group Ⅲ n = 8). Group Ⅰ was operated but without ischemia reperfusion. Group Ⅱ was ischemia reperfusion. And Group Ⅲ was received TSN. M J Smith' s ischemia model blood was withdrawn until the mean pressure reached 50 mmHg, via the right femoral arterial catheter. Clamps were removed after thirty minutes ischemia. Thereafter,reperfusion had been performed for four hrs. Group Ⅲ was administrated TSN 3 mg. Cerebral tissue EB content Cytosolic Ca^2+ and water content were recorded and measured. Results Cellular isolated calcium and water content in Group Ⅱ and Group Ⅲ were higher than that in the sham group, whereas that in Group Ⅲ were significantly lower than that in Group Ⅱ statistically. Conclusion TSN could protect the ischemia b rain and decrease blood brain barrier.
