摘要 目的:在胚鼠中脑曲分离神经干细胞,探讨IL-1α诱导胚鼠中脑神经干细胞分化为多巴胺神经元的效能。方法:体外分离、培养小鼠胚胎的中脑神经干细胞,加入IL-1α诱导其分化,通过巢蛋白(Nes-tin)、神经元特异性烯醇化酶(NSE)、胶质纤维酸性蛋白(GFAP)、酪氨酸羟化酶(TH)免疫组化鉴定,并统计分析其诱导效能。结果:胚鼠中脑的腹侧中脑曲部位分离获得了大量未分化、呈巢状悬浮生长的细胞球,且能分化为神经元和神经胶质细胞;IL-1α能明显提高NSC定向分化为多巴胺能神经元比例(IL-1α组14.3%±0.8%vs对照组2.6%±0.3%,P<0.05)。结论:胚鼠中脑存在有多向分化潜能的神经干细胞和中脑神经干细胞被IL-1α诱导向多巴胺能神经元的分化比例明显升高。 Objective:To study the effect of Interleukin-1αon the differentiation of mese-ncephalic neural stem cells(NSCs) of embryo mice-derived which were isolated from embryonic mice into dopaminergic neurons in vitro.Methods: Mesencephalic NSCs were isolated from emoryonic mice mesencephalon and cultured in vitro.After seven days,these cells were.induced to differentiate by IL-1α.The differentiated cells were identified by immunocytochemical staining with nestin,neuro-specific enolase(NSE),glial fibrillary acidic protein (GFAP),tyroxine hydroxylase(TH).The detected cells were analyzed by cell count.Results:Clusters of neural stem cells were obtained in the supernatant and the cells could be differentiated into neurons and astrocytes,About 14.3%±0.4%(P<0.05) of the neural stem cells were induced into dopaminergic neurons by IL-1α.Conclusion:Neural stem cells from ventral mesencephalon can be cultured、passaged and differentiated naturally.differentiation into dopaminergic neurons was significantly rising in presence of(IL-1α)
