摘要
目的研究豚鼠视网膜电图的特点及记录方法.方法采用RETI-port系统,自制的银-氯化银环形角膜记录电极和不锈钢针状电极,依据国际电生理协会所制定的常规5项标准,记录10只成年豚鼠视网膜电图的5种基本反应:视杆细胞反应(Rod-ERG)、暗适应眼的最大混合反应(MaX-ERG)、振荡电位(OPs)、视锥细胞反应(Cone-ERG)与对快速重复刺激的反应(Flick ERG).结果豚鼠Rod ERG波形与人类和大鼠Rod-ERG相似,但是b波幅值相对较低约为(97.98±8.76)μV.Max-ERG a波幅值下降明显,大概(119.08±12.92)μV.Max-ERG b波相对平缓,无陡峭尖峰,呈缓慢上升趋势,峰时较为靠后,潜伏期(85.30±3.18)ms,幅值(115.93±12.28)μV.豚鼠OPs波一般为3~4个波,其中第2个波(O2)、第3个波(O3)和第4个波(O4)幅值接近,以O2波幅值最高(29.00±3.07)μV.Cone-ERG a波与正常大鼠稍有不同,a波下降较为明显,幅值(17.38±2.27)μV,b波幅值较正常大鼠稍低,约为(47.52±4.08)μV.Flick-ERG的波形在刺激频率为15 Hz时最为稳定,固定为4.5个波,波形平滑,幅值(31.71±4.76)μV.结论正常豚鼠视网膜电图与大鼠和人类具有一定的相似性,同时又具有自己的特点.我实验室所建立的小动物视网膜电图记录方案能够准确、可靠的记录豚鼠视网膜电图,为进一步研究豚鼠视网膜电图的特点奠定了一定基础.
Objective To investigate the recording condition and characteristics of normal electroretinogram(ERG)in guinea pig.Methods ERGs of 10 normal guinea pigs ( 10 eyes)were measured following the standards of International Society for Clinical Electrophysiology of Vision including Rod-ERG, Max-ERG,OPs, Cone-ERG and Flick-EFG.With RETI-port system and homemade silver-silver chloried ring form cornea recording electrode and stainless steel needle electorde. Results The Rod-ERG of guinea pig was similar to that of the normal rat and human being in some content, ttowever, the amplitude of b-wave was relatively lower with the value of about(97.98±8.76)μV. The reduction of the amplitude of a-wave in Max-ERG was more distinctive while the reduction of b-wave was relatively gentle. The amplitude of a-wave in Max-ERG was about ( 119.08 ± 12.92 ) μV, and b-wave was ( 115.93 ± 12.28 ) μV. The implicit time of b-wave was(85.30 ± 3.18)ms. There was no steep peak of b-wave in Max-ERG. However it shows a tendency to rise slowly and the peak time was delayed. The wave of OPs always had 3 to 4 waves. The amplitudes of O2,O3 and 04 were close to each other,and O2 was often the highest among them. The amplitude of O2 was(29.00 ± 3.07) μV. The amplitude of a-wave of Cone-ERG decreased obviously with the value of( 17.38 ± 2.27) μV, while bwave decreased gently. The amplitude of b-wave was (47.52 ± 4.08)μV. The waves of Flick-ERG always had 4.5 waves and were more stable at 15 Hz. The amplitude of which was(31.71 ± 4.76)μV.Conclusion The electroretinogram of guinea pig is similar to rat and human being,however it also has some own characteristics. The recording procedure founded by our lab can evaluate the electroretinogram of guinea pig reliably.
