摘要
研究了一株芽孢杆菌(BacillusSp.)产生脲酶的条件及脲酶的提取。结果表明,芽孢杆菌适宜的生长及产酶温度为35~37℃,产酶受底物──尿素的诱导。静止培养有利于产酶,Ni2+对产酶没有影响。脲酶粗提物经硫酸铰盐析、热处理、SephadexG—200及DEAE-纤维素柱层析等提取步骤脲酶比活力提高了近70倍,回收率15.5%。
The paper describes the experimental results of production and extraction of ureasefrom Bacillus sp The results showed that the strain was suitable for temperature of growth and ureaseproduction at 35-37℃, the enzyme production was induced by its substrate-urea, static culturedoes good to enzyme yield,but Ni2+ addition has no effect on it.After some extraction steps such asammonium sulfate fractionation, heat treatment and column chromatography on Sephadex G-200 andDEAE-cellulose,enzyme specific activity was increased about 70 times compared with initial product,the rate if recovery was 15.5%.
