摘要 目的:观察玉竹多糖对D-半乳糖致亚急性衰老模型小鼠血清中抗氧化系统超氧化物歧化酶活性和丙二醛含量的影响,探讨玉竹多糖抗衰老的可能作用机理,为中药延缓衰老的研究开辟新的领域。方法:实验于2004-07/2005-05锦州医学院免疫教研室完成。取昆明小鼠50只,随机分正常对照组、衰老模型组和0.5g/kg,1g/kg,2g/kg的玉竹多糖治疗组,共5组。除正常对照组外,其余各组每只颈背部皮下注射50g/LD-半乳糖0.5mL,连续6周。同时治疗组腹腔分别注射0.5g/kg,1g/kg,2g/kg3种不同剂量的玉竹多糖给予治疗,正常对照组和衰老模型组每只腹腔注射生理盐水0.5mL。6周后,麻醉下将小鼠摘除眼球取血,常规分离血清,在4℃下3000r/min离心5min,取上清待测。用分光光度计检测小鼠血清中超氧化物歧化酶活性和丙二醛含量,并将小鼠称质量并剖取小鼠胸腺和脾脏,用电子天平称质量,以脏器湿质量(mg/g)表示小鼠脾指数和胸腺指数,观察其对免疫功能的影响。结果:50只实验动物数据全部进入结果分析。①衰老模型组与正常对照组比较,血清中超氧化物歧化酶活性显著降低,丙二醛水平明显升高[(151.39±41.74),(338.39±32.42)μU/L;(13.56±2.06),(5.36±0.47)μmol/L;F=3.239,P<0.05];胸腺指数和脾指数下降[(2.19±0.90),(3.86±0.63)mg/g;(4.16±1.42),(6.48±1.49)mg/g;]。②治疗组与衰老模型组比较,玉竹多糖2g/kg剂量组能显著提高衰老模型小鼠血清中超氧化物歧化酶活性,降低丙二醛含量[(443.21±42.70)μU/L,(5.55±0.88)μmol/L;F=4.727,P<0.01或F=3.254,P<0.05]。3种不同剂量的玉竹多糖均能提高脾指数,但3个治疗组之间的差异无统计学意义。结论:玉竹多糖具有抗氧化作用,对免疫功能有一定的调节作用。玉竹多糖可能通过提高超氧化物歧化酶活性,增强其对自由基的清除能力,抑制脂质过氧化,降低丙二醛含量,从而减轻对机体组织的损伤以延缓衰老。 AIM: To study the anti-aging effect of polygonatum odoratum polysaccharide on the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) of rats with subacute senile model by Dgalactose, and research the effect of polygonatum odoratum polysaccharide on anti-aging so as to provide the new field for Chinese herb. METHODS: The experiment was completed in the Department of Immunology of Jinzhou Medical University from July 2004 to May 2005. Fifty kunming mice were randomly divided into five groups: normal control group, senile model group and 3 test groups with 0.5 g/kg, 1 g/kg and 2 g/kg of polygonatum odoratum polysaccharide. Nap of mice were injected with 50 g/L 0.5 mL D-galactose except normal control group for six successive weeks; meanwhile, mice in treatment group were injected with 0.5 g/kg, 1 g/kg and 2 g/kg polygonatum odoratum polysaccharide respectively, and mice in the normal control group and the senile model group were injected with 0.5 mL saline. Six weeks later, eyeball of mice was taken out at anesthesia stage; serum was separated routinely and centrifugated at 3 000 r/minute for 5 minutes at 4℃. The upper serum was obtained for measurement. The activity of SOD and contents of MDA in serum of the mice were determined with speetrophotometer. Meanwhile, mice were weighted and thymus and spleen were taken out to weight with electrical balance. Wet mass of organs was used to express indexes of thymus and spleen so as to observe the effect on immunological function. RESULTS: All 50 mice entered the final analysis. (1) Compare the senile model group with normal control group, the activity of SOD decreased significantly and contents of MDA in serum increased significantly [(151.39±41.74), (338.39±32.42)μU/L; (13.56±2.06), (5.36±0.47)μmol/L; F=3.239, P〈0.05]; indexes of thymus and spleen were decreased [(2.19±0.90), (3.86±0.63)mg/g; (4.16±1.42), (6.48±1.49) mg/g]. (2) Compare treatment group with senile model group, the 2 g/kg dose of polygonatum odoratum polysaccharide could increase SOD activity and decrease MDA content of the serum in subaeute aging model mice [(443.21±42.70) μU/L, (5.55±0.88)μmol/L; F=4.727, P〈0.01 or F=3.254, P 〈 0.05]. It could also increase spleen index by three doses of polygonatum odoratum polysacchadde, but differences among three groups were not significant. CONCLUSION: Polygonatum odoratum polysaccharide has antioxidation action. It has certain action to regulate immune function. Polygonatum odoratum polysaccharide have the effect on anti-aging by improving the aetivity of SOD, strengthening the activity Of taking away free radicals, inhibiting lipid peroxidation and reducing the contents of MDA.
